A postdoctoral position has arisen in the lab of Dr Florian Hollfelder, University of Cambridge, Department of Biochemistry, Cambridge and is available for 12 months from 1st April 2014 on evolution and ancestral reconstruction of highly promiscuous enzymes (namely members of the alkaline phosphatase that catalyse up to 6 chemically distinct activities). We will base directed evolution of new generalists and specialists on measurement of fitness landscapes of randomised protein populations: such distribution functions for multiple activities will be gathered by ultrahigh-throughput screening of >106 clones in picolitre droplet compartments during in vitro and in vivo evolution cycles, followed by systematic characterization of multiple fitness traits such as expression level, stability or reaction kinetics. These approaches will enable a dynamic picture of protein evolution at various levels from transition state recognition, protein biophysics to population genetics. The project involves collaboration with the groups of Erich Bornberg-Bauer at M?nster (http://ieb.uni-muenster.de/bioinf/people/ebb) and Nobu Tokuriki at Vancouver (http://www.msl.ubc.ca/faculty/tokuriki).

Applicants must have a PhD or a equivalent and should ideally have a strong background in protein engineering, chemical biology, enzyme kinetics, molecular biology, enzyme mechanism.

Fixed-term: The funds for this post are available for up to 12 months in the first instance.
