  PhD student / Postdoc Position in computational analysis and modelling of alternative splicing decisions

  The Institute of Molecular Biology gGmbH (IMB) is a Centre of Excellence for Life Sciences, funded by the Boehringer Ingelheim Foundation. It is located within the campus of the Johannes Gutenberg University, Mainz, Germany. Applications are invited for a Post-Doctoral Research Fellows to study the molecular mechanisms of alternative splicing in the research group of Dr Stefan Legewie.   Project

  We aim for a better understanding of splice-regulatory mechanisms using quantitative systems biology approaches. Along the lines of our previous research, high-throughput mutagenesis data will be used to comprehensively define cis-regulatory elements controlling certain disease-relevant splicing events (see Braun et al., 2018). These data will be theoretically analysed using a combination of bioinformatics, machine learning and kinetic modelling approaches. This way, we will gain insights into functional roles single-nucleotide polymorphisms in human populations. Furthermore, we hope to better understand kinetic proofreading mechanisms that promote the fidelity of splicing decisions, while at the same time allowing for flexible control of alternative splicing. The role of regulatory proteins controlling alternative splicing will be investigated using a combination of bioinformatics, modelling and perturbation experiments (see Sutandy et al., 2018, Braun et al., 2018). Our mechanistic mathematical models of splicing regulation will be employed to better understand gene expression at the single-cell level, where transcription and alternative splicing are stochastic processes. Using single-cell RNA sequencing data, we will study heterogeneity in isoforms, discriminate possible origins of splicing stochasticity, and investigate the potential biological significance of noisy splicing in development and disease. These data will be interpreted using quantitative stochastic models of transcription and splicing (see Fritzsch et al., 2018).

  You have

  ·         MSc / PhD in Bioinformatics, Computational Biology, Natural Sciences or related areas

  ·         Sound experience in large-scale data analysis (RNA-Seq) and/or mathematical modelling of biological networks

  ·         Excellent communication skills and good team spirit with the ability to solve problems independently

  ·         Fluent in English (spoken and written)

  The Medical Research Council Toxicology Unit is an internationally renowned institution focused on the delivery of field-changing mechanistic insights into toxicology and disease. The Unit is equipped with state-of-the-art facilities including Cryo-EM and offers excellent opportunities for postdoctoral development. The Unit is part of the University of Cambridge; it is currently located at Leicester and will physically relocate to Cambridge in 2020.

  We are looking to appoint an MRC Postdoctoral Fellow (PF) to work in the laboratory of Prof. Marion MacFarlane to carry out research into the 'Molecular Mechanisms of Cell Death'.

  Our laboratory is focused on investigating cell death pathways that are regulated in response to members of the TNF cytokine family and agents that trigger mitochondrial-dependent cell death, with a specific emphasis on mechanisms that regulate activation of cell death signalling complexes.

  The aim of the project is to obtain novel insights on the regulation and 3D molecular architecture of key multiprotein signalling complexes that direct cell death/survival.

  This is a three year training and development position during which you will receive a range of formal and on-the-job training to ensure you receive the skills relevant to your role and to help support your future career.

  Applicants must have (or be in the final stages of receiving) a PhD in a relevant discipline and have experience in protein expression/the analysis of protein structure and function. Experience in Cryo-Electron Microscopy/analysis is desirable. The ability to publish high quality research in a peer reviewed journal; to be able to work both independently and as part of a team, together with excellent communication and problem solving skills are required.

  This post requires you to hold a PhD in a relevant discipline.
